If you keep one card in the internal slot you can use the 'backup/restore' function in the 'tools' menu (click the application list). This is useful for when you upgrade, particularly in case you have already configured your N800 (e.g. bluetooth pairing with phone, email setup etc.)

I downloaded the nokia software update wizard and installed it.
I connected to the device and the software tells me that the DEVICE NOT FOUND and it asked me to hold the HOME key and switch on the Power button till the usb logo appears on top right of the screen. I did exactly what they said but the software didnt react , still shows device not found in my pc. While Nokia shows the usb logo top right and NOKIA logo on screen.
Nothing happened. When i removed the usb then device boots normally.
Instead of pressing the HOME key and all that tricky magic, you should instead: 1) Remove charger cable from N800 2) connect, and start software wizard, then 3) Now just plug in the charger cable. It will do the same trick as the HOME/power button magic, but it's much simpler. No buttons to press.

They all told me to add the repositiory using Application manager. But now the problem is there is no provision to add new repositiory links in the appmanager.
- Start application manager
- In the upper left corner there's a drop-down menu (a little down arrow under the title).
- Click it, find 'Tools', the third item down will be the application catalogue (or whatever its equivalent in your language locale).
- Here you can add repositories. An excellent list of repositories can be found here:
http://www.gronmayer.com/n800/repos/index.php?lang=en
(NB: It's unlikely you'll need all of them. Those including 'repository.maemo.org' and 'bora' are those you want first. And those with 'nokia.com' in the name - note that there's probably one or more of those pre-configured already)

EDIT: Added the bit about nokia.com repositories
